Hydroxymycotrienin A
Hydroxymycotrienin A is an ansamycin group antibiotic with anticancer effects. Hydroxymycotrienin A suppresses integrated HPV18 E6 and E7 gene expression. Hydroxymycotrienin A inhibits growth of HPV-positive cervical cancer cells, stabilizes conformational mutant p53R175H and contact mutant p53R273H via non-covalent interactions, and acts as a non-covalent pharmacological chaperone. Hydroxymycotrienin A can be used for the research of human cervical cancer and non-small cell lung cancer (NSCLC).

Hydroxymycotrienin A potently inhibits the growth of HPV-positive human cervical cancer cell lines (SiHa, CaSki, HeLa) with IC50 values ranging from 0.07 to 0.18 μg/mL, and is 3-10 times more potent against these lines than against HPV-negative C33A and HT-3 cells[1].
Hydroxymycotrienin A (1 μg/mL; 24 h) inhibits HPV18 E6/E7 gene expression in HeLa cells without significantly altering the expression of a host control gene[1].
Hydroxymycotrienin A (100 μg/mL) shows no antimicrobial activity against tested bacteria, fungi, or yeast[1].
Hydroxymycotrienin A binds human native p53, p53R175H mutant, and p53R273H mutant with high non-covalent affinities (-5.87 kcal/mol, -6.63 kcal/mol, and -6.57 kcal/mol, respectively), outperforming APR-246 in all docking scenarios[2].
Hydroxymycotrienin A (500 ns) acts as a structural stabilizer for human native p53, p53R175H, and p53R273H over 500 ns molecular dynamics simulations, reducing backbone deviations, residue fluctuations, and solvent exposure while preserving structural compactness and maintaining stable hydrogen bond interactions[2].
