Bethoxazin is a yeast DNA topoisomerase II inhibitor with an IC50 of 5.3 μM, and also a broad-spectrum industrial microbicide and covalent adduct-forming agent. Bethoxazin inhibits the catalytic activity of yeast DNA topoisomerase II by reacting with the key free cysteine thiol group on the enzyme. Bethoxazin forms covalent adducts with molecules containing free thiol groups. Bethoxazin inhibits the growth of yeast cells. Bethoxazin can be used in studies related to the growth of fungi, molds and algae.

Biological Activity
Bethoxazin (30-100 μM) is highly reactive toward free sulfhydryl groups (GSH, cysteine, HSA) but does not react with amino, carboxylic, phenolic, amino oxo, alcoholic, or phosphate functional groups under tested conditions[1].
Bethoxazin (100 μM; 8 h) undergoes nucleophilic attack at C3 of its oxathiazine oxide ring by sulfhydryl groups, forming ring-opened intermediates that react with a second sulfhydryl molecule to produce disulfide adducts[1].
Bethoxazin (66 μM; 5 h) forms a covalent adduct with free human serum albumin at an equimolar ratio, reacting with the single free cysteine residue of HSA[1].
Bethoxazin (0.01-100 μM; 6 h) potently inhibits the growth of yeast BY4742 cells with an IC50 of 11 μM[1].
