Peanut Agglutinin (PNA) Biotinylated is a biotinylated lectin that specifically recognizes and binds to the Galβ1-3GalNAc sequence, thereby targeting SPESP1 and TF blood group antigens. As an acrosome-specific marker, Peanut Agglutinin Biotinylated stains mouse spermatocytes and sperm acrosomes with high specificity, and this binding depends not only on SPESP1 but also recognizes other testicular acrosomal glycoproteins. Peanut Agglutinin Biotinylated accurately identifies TF antigens in Reed-Sternberg cells, histiocytosis X cells, and esophageal squamous cell carcinoma, without cross-reacting with non-specific cells. Peanut Agglutinin Biotinylated maintains staining stability in both B-5 and formalin fixatives, and serves as an important tool for the study of Hodgkin's disease, histiocytosis X, and various malignant tumors.

Peanut Agglutinin Biotinylated (4 µg; 30 min) specifically binds to sperm equatorial segment protein 1 as its primary glycoprotein target in wild-type mouse testis lysate, with SPESP1 detected in all six purification and LC-MS/MS analyses[1].
Peanut Agglutinin Biotinylated (10 μM/mL; 30 minutes plus 30 minutes with mouse liver powder) selectively stains histiocytosis X cells and interdigitating reticulum cells with a unique surface and paranuclear pattern, distinguishes these cells from benign macrophage-histiocytes via staining pattern differences, and fails to produce a signal in malignant histiocytosis and monoblastic leukemia cells, enabling differential diagnosis in paraffin-embedded human tissues[3].
Biotinylated Peanut Agglutinin (Biotinylated) (5 µg/mL; 1 h) detects significantly elevated circulating TF-antigen (mean absorbance 0.319-0.411) in pre- and post-treatment well, moderately, and poorly differentiated human esophageal squamous cell carcinoma patients, with an overall sensitivity of 87.5% and specificity of 90%[4].
Peanut Agglutinin Biotinylated detects TF-antigen expression in all human esophageal squamous cell carcinoma specimens, with intense expression present in 52.0% of moderately differentiated and 54.55% of poorly differentiated ESCC specimens, compared to only 8.70% of well differentiated ESCC specimens[4].
