Biological Activity
DPP9 Protein, Human (sf9) is the recombinant human-derived DPP9, expressed by sf9 insect cells , with tag Free labeled tag.
DPP9 is a dipeptidyl peptidase that cleaves N-terminal dipeptides from proteins with Pro or Ala at position 2. It acts as a key inhibitor of caspase-1-dependent pyroptosis in monocytes and macrophages by preventing NLRP1 and CARD8 activation in resting cells. DPP9 sequesters the active C-terminal fragments of NLRP1 and CARD8 inflammasomes in a ternary complex, blocking their oligomerization and activation. While its dipeptidyl peptidase activity is required for NLRP1/CARD8 suppression, neither protein is a direct substrate, implying the involvement of unidentified substrate(s). by similar: Its function parallels DPP8 but may involve distinct mechanisms.
In the activity assay, DPP9 acted on the non-fluorescent substrate Gly-Pro-PNA and cut it to release free p-nitroaniline (PNA). The free PNA with characteristic absorption at 405nm was detected by colorimetry with a specific activity of 2628 U/μg.
