IGF-I R Protein, Mouse (HEK293, His)

Cat. No.: HY-P78723
COA Handling Instructions

The IGF-I R protein, a receptor tyrosine kinase, mediates insulin-like growth factor 1 (IGF1) actions, with high affinity for IGF1. Upon ligand binding, IGF1R activates, leading to autophosphorylation and tyrosine phosphorylation of substrates, including IRS1/2, Shc, and 14-3-3 proteins. Phosphorylation initiates PI3K-AKT/PKB and Ras-MAPK pathways, regulating proliferation and apoptosis. IGF1R signals through JAK/STAT, activating STAT3, contributing to its transforming activity. It also activates JNK and inhibits JNK activation, forming a hybrid receptor with INSR to bind IGF1. IGF-I R Protein, Mouse (HEK293, His) is the recombinant mouse-derived IGF-I R protein, expressed by HEK293, with C-His labeled tag. The total length of IGF-I R Protein, Mouse (HEK293, His) is 906 a.a., with molecular weight of 110-140 & 50-65 kDa, respectively.

Background

The IGF-I R protein, a receptor tyrosine kinase, serves as a mediator for the actions of insulin-like growth factor 1 (IGF1). It exhibits a high affinity for IGF1, lower affinity for IGF2, and insulin (INS). Activation of IGF1R plays a pivotal role in controlling cell growth and survival, with critical implications in tumor transformation and the survival of malignant cells. Upon ligand binding, the receptor kinase is activated, leading to autophosphorylation and the tyrosine phosphorylation of various substrates, including insulin-receptor substrates (IRS1/2), Shc, and 14-3-3 proteins. Phosphorylation of IRS proteins initiates two main signaling pathways: the PI3K-AKT/PKB pathway and the Ras-MAPK pathway. Activation of the MAPK pathway enhances cellular proliferation, while activation of the PI3K pathway inhibits apoptosis and stimulates protein synthesis. Furthermore, IGF1R signals through the Janus kinase/signal transducer and activator of transcription pathway (JAK/STAT), where phosphorylation of JAK proteins can activate STAT proteins, particularly STAT3, contributing to the transforming activity of IGF1R. Additionally, IGF1R activates JNK kinases and exerts inhibitory effects on JNK activation. When forming a hybrid receptor with INSR, IGF1R binds IGF1.

Biological Activity

1. Immobilized Mouse IGF1R, His Tag at 1 μg/mL (100 μl/well) on the plate. Dose response curve for Human IGFI, hFc Tag with the EC50 of 2.58 μg/mL determined by ELISA.
2.Measured by its binding ability in a functional ELISA. When Recombinant Mouse IGF-I R is immobilized at 5 µg/mL (100 µL/well) can bind Recombinant Human IGF-I. The ED50 for this effect is 322.3 ng/mL.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

  • Specific Activity Calculator

The reconstitution calculator equation

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration
= ÷

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×
C1   V1   C2   V2

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
Unit/mg = 106 ÷ ng/mL
