Background

MT-RNR1 regulates insulin sensitivity and metabolic homeostasis by inhibiting the folate cycle, reducing de novo purine biosynthesis, and accumulating the intermediate AICAR to activate AMPK. It protects against age-related and diet-induced insulin resistance and obesity. Under metabolic stress, it translocates to the nucleus, binds to AREs, and modulates gene expression in an NFE2L2-dependent manner to enhance cellular stress resistance. Additionally, it increases mitochondrial respiration and levels of CPT1A and cytokines (IL1B, IL6, IL8, IL10, TNF) in senescent cells. MT-RNR1 enhances mTORC2 activity while suppressing PTEN, promoting AKT phosphorylation and subsequent FOXO1 inhibition, reducing MSTN levels to stimulate skeletal muscle growth. Via the TGFB/SMAD pathway, it drives osteogenic differentiation of bone marrow mesenchymal stem cells and boosts osteoblast proliferation and type I collagen (COL1A1, COL1A2) synthesis.
