TMEM175 is a proton-activated proton channel that facilitates proton efflux from endosomes and lysosomes to maintain steady-state pH. It is activated at low pH (below 4.6) and selectively mediates lysosomal proton release, balancing V-ATPase activity for pH homeostasis (by similar: PubMed:35750034). Additionally, TMEM175 functions as a potassium channel at higher pH, regulating potassium conductance in endosomes and lysosomes. It serves as the pore-forming subunit of the lysoK(GF) complex, which consists of TMEM175 and AKT (AKT1, AKT2, or AKT3) and is activated by extracellular growth factors. The lysoK(GF) complex opens the TMEM175 channel via AKT-induced conformational changes, activating its function and playing a critical role in protecting neurons from stress-induced damage (by similar: PubMed:33505021).
