C-TPA is a fluorescent probe used for hypochlorite detection and bioimaging of lipid droplet polarity in cancer cells. For hypochlorite detection, when its derivative C-TPA-S is exposed to hypochlorite, an oxidative desulfurization reaction converts the thiocarbonyl group of C-TPA-S to the carbonyl group of C-TPA, thus restoring bright fluorescence from the almost non-fluorescent C-TPA-S. For bioimaging of lipid droplet polarity, the intramolecular charge transfer process between its Triphenylamine (HY-W011998) donor group and Coumarin (HY-N0709) acceptor group endows C-TPA with solvatochromism-it exhibits stronger fluorescence in less polar environments (such as lipid droplets in cancer cells) and weaker fluorescence in more polar environments (such as lipid droplets in normal cells), enabling the differentiation of cancer cells and cancer tissues from normal cells and normal tissues. The excitation wavelength of C-TPA is 405 nm, and the emission wavelengths used for cell and tissue imaging cover 425-475 nm and 500-550 nm; in solvents, its emission peak shifts from 494 nm in non-polar toluene to 528 nm in polar water .

ZnPc-PEG2-VH032 is a VHL-pathway-dependent photodegradation targeting chimera (PDTAC) and cytotoxic agent. ZnPc-PEG2-VH032 (HY-120217) binds to the VHL ligand domain, and then specifically degrades VHL under light irradiation, a process independent of non-specific ROS-mediated protein damage. ZnPc-PEG2-VH032 uses Zinc phthalocyanine (HY-19204) as a photosensitizer, and generates ROS via type I and type II photodynamic pathways under 680 nm LED irradiation. On one hand, it targets and degrades the bound VHL protein through ROS; on the other hand, it exerts direct photodynamic cytotoxicity. Meanwhile, the degradation of VHL downregulates the phosphorylation level of CDK2/4, induces cell cycle arrest in tumor cells, further enhances the sensitivity of tumor cells to oxidative damage caused by ROS, and achieves a synergistic anti-tumor effect. Oxygen homeostasis regulation is the most fundamental cellular process for adjusting physiological oxygen variations, and its irregularity leads to various human diseases, including cancer. Hypoxia is closely associated with cancer development, and hypoxia/oxygen-sensing signaling plays critical roles in the modulation of cancer progression.

Hypoxia-inducible factor 1 (HIF-1) is a transcription factor that functions as a master regulator of oxygen homeostasis. A variety of HF-1 target genes have been identified thus far which encode proteins that play key roles in critical developmental and physiological processes including angiogenesis/vascular remodeling, erythropoiesis, glucose transport, glycolysis, iron transport, and cell proliferation/survival.

HIF-1 is a heterodimeric transcription factor consisting of a constitutively expressed β-subunit and an oxygen-regulated α-subunit. The unique feature of HIF-1 is the regulation of HIF-1α expression and activity based upon the cellular O2 concentration. Under normoxic conditions, hydroxylation of HIF-1α on these different proline residues is essential for HIF proteolytic degradation by promoting interaction with the von Hippel-Lindau tumor-suppressor protein (pVHL) through hydrogen bonding to the hydroxyproline-binding pocket in the pVHL β-domain. As oxygen levels decrease, hydroxylation of HIF decreases; HIF-1α then no longer binds pVHL, and becomes stabilized, allowing more of the protein to translocate to the cell’s nucleus, where it acts as a transcription factor, upregulating (often within minutes) the production of proteins that stimulate blood perfusion in tissues and thus tissue oxygenation.
