Endosidin-2 (ES2) is a selective inhibitor targeting the conserved exosome subunit EXO70. Endosidin-2 binds to the C-terminal domain of EXO70, inhibiting exocytosis and endosomal recycling, while promoting vacuolar trafficking in plant cells. Endosidin-2 interferes with the EXO70-mediated vesicle-to-plasma membrane anchoring process, leading to abnormal aggregation of auxin transporters (such as PIN2) in the cytoplasm and redirected to vacuolar degradation, while causing abnormal Golgi structure (such as cup-shaped or ring-shaped vesicles cisternae formation). Endosidin-2 can inhibit exocytosis in plant and mammalian cells and is mainly used to study the dynamic regulation of membrane trafficking (such as polar growth, vesicle sorting) .

NPPM 6748-481 is a potent and selective yeast Sec14 inhibitor with a yeast Sec14 IC50 of 211 nM. NPPM 6748-481 disrupts membrane trafficking, impairing trans-Golgi network-endosomal transport and endocytic transport. NPPM 6748-481 impairs Snc1 v-SNARE recycling, secretory invertase trafficking, and vacuolar carboxypeptidase Y processing. NPPM 6748-481 can be used for research on yeast infection .

CMLD009688 is a cationic amphiphilic antifungal agent. CMLD009688 selectively inhibits plant pathogenic fungi such as Fusarium graminearum. CMLD009688 interacts with biological membranes, perturbing vacuolar and mitochondrial membrane structures to induce fungal cell death. CMLD009688 is promising for research of plant fungal diseases (e.g., wheat head blight, gray mold) .

IPA is a specific Sec18/NSF inhibitor. IPA binds to phosphatidic acid competitively, induces conformational rigidification of Sec18/NSF, disrupts its essential flexibility, and blocks SNARE priming and vacuolar membrane fusion. IPA can be used to investigate the activity of Sec18 and SNARE priming kinetics .

AM4-64 is a fixable cationic styryl fluorescent dye for nerve terminals, which can be used for vesicle cycle evaluation, endocytic pathway tracing and endocytosis detection. AM4-64 inserts into the plasma membrane, is internalized via activity- or energy-dependent endocytosis, and accumulates in vesicles, endosomal compartments, vacuolar membranes, mitochondria and endolysosomal structures. The fluorescence of AM4-64 is enhanced in the membrane-bound state. AM4-64 is applicable to relevant studies on neurons, plant and fungal cells and tissues (Ex/Em = 510/750 nm) .

NIK-12192 is an orally active inhibitor of vacuolar H +-ATPase. NIK-12192 reduces intracellular pH, decreases lysosomal volume and acidity, and alters the intracellular localization of V-ATPase by inhibiting proton pumps . NIK-12192 induces αvβ5 integrin polarization, cytoskeletal disruption, cell detachment, anoikis-mediated delayed apoptosis and necrosis, a delayed reduction in mitochondrial membrane potential, and lysosomal/phagosomal accumulation. NIK-12192 inhibits tumor cell migration, invasion, and three-dimensional spheroid growth, and enhances the activity of Topotecan (HY-13768). NIK-12192 suppresses spontaneous lung metastasis in vivo. NIK-12192 is used in research related to colon cancer, ovarian cancer, lung cancer, breast cancer, renal cancer, prostate cancer, acute myeloid leukemia, and melanoma .
