3-Methylbenzene-1,2-diamine (Tolylene-2,3-diamine) is a CD44 hyaluronic acid-binding domain ligand, with a Kd of 8.5 mM for hCD44 HABD and a Kd of 6.4 mM for mCD44 HABD. 3-Methylbenzene-1,2-diamine acts as a non-inhibitor that does not interfere with the binding of CD44 HABD to polymeric hyaluronic acid in vitro.

3-Methylbenzene-1,2-diamine (Compound 1) binds to the recombinant hCD44 hyaluronic acid-binding domain, with a KD value of 8.5 mM[1].
3-Methylbenzene-1,2-diamine binds to the recombinant mouse CD44 hyaluronic acid-binding domain with a Kd value of 6.4 mM, occupies adjacent subsites of the HA-binding groove, and induces specific protein conformational changes[1].
3-Methylbenzene-1,2-diamine does not inhibit the binding of recombinant hCD44 hyaluronic acid-binding domain to immobilized polymeric HA in vitro[1].
