4-Chloro-N-phenylpyrimidin-2-amine (compound 4) is synthesized via chlorination of 2-anilinopyrimidin-4 (5H)-one using phosphorus oxychloride at 100 ℃, and is isolated as a brown solid. This compound serves as a key intermediate for Suzuki coupling, and can react with indole boronic acid derivatives followed by deprotection to prepare the target pyrimidine-indole derivatives[1].
