Acetyl-cyclosporin A aldehyde is an acetylated derivative of Cyclosporin A (HY-B0579) with a reducible aldehyde group. Acetyl-cyclosporin A aldehyde serves as a synthetic intermediate for the calcineurin inhibitor Voclosporin (HY-106638). Acetyl-cyclosporin A aldehyde can act as a substrate for ruthenium nanocatalyzed reductive deuteration reactions, producing the corresponding deuterated alcohol with selective deuterium incorporation at the α-position of the oxygen atom.

Acetyl-cyclosporin A aldehyde is prepared from acetylcyclosporin A via ozonolysis, followed by conversion to Voclosporin through the Peterson reaction/hydrolysis[1].
Acetyl-cyclosporin A aldehyde undergoes regioselective reductive deuteration at the α-position of the oxygen atom under 1 bar D2 atmosphere, producing 87% deuterated alcohol[2].
