Background

  • Function

DX39A is a helicase that plays a key role in mRNA export, participating in multiple steps of RNA metabolism, including alternative splicing. DX39A regulates the export of nuclear mRNA to the cytoplasm by binding to its extracellular domain (ECD). Furthermore, it participates in the export of uridine-rich small nuclear RNA (U snRNA) from the spliceosome by stimulating the binding of the adaptor protein PHAX to RNA. DX39A also exerts antiviral effects independently of the interferon pathway by binding to conserved 5' sequence elements in viral genomic RNA[1]>[2].

  • Subcellular Localization

    Nucleus, Cytoplasm

  • Expression

Tissue specificity: Detected in testis, and at lower levels in brain, kidney, lung, thymus, spleen and salivary gland.
Induction: Up-regulated in proliferating cells. Present at low levels in quiescent cells.

  • Isoforms & Post-Translational Modification

DDX39A has three isomers: O00148-1: 427 amino acids, molecular weight 49,130 Da (predicted); O00148-2: 322 amino acids, molecular weight 36,577 Da (predicted); O00148-3: 267 amino acids, molecular weight 30,488 Da (predicted).
SUMOylation occurs after RANBP2 modification; SUMOylation affects its binding ability to RNA.

  • Subunit

Binds ALYREF/THOC4 and DDX39B/BAT1. Interacts with the apo-AREX complex component SARNP. Interacts with MX1. Interacts with MCM3AP isoform GANP. Interacts with ECD. Interacts with PHAX; this interaction stimulates PHAX RNA binding activity.
