Immunohistochemical analysis of paraffin-embedded human Colon cancer tissue using uPAR antibody was performed. The section was pretreated using high-temperature and high-pressure mediated EDTA antigen retrieval buffer (pH 9.0), for 5 minutes. The tissues were incubated with primary antibody (HY-P81512, 1:100 dilution) at room temperature for 60 minutes. Detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. The tissues were counterstained with hematoxylin and mounted with neutral balsam mounting medium.

Background

  • Function

uPAR acts as a receptor for urokinase plasminogen activator. Plays a role in localizing and promoting plasmin formation. Mediates the proteolysis-independent signal transduction activation effects of U-PA. It is subject to negative-feedback regulation by U-PA which cleaves it into an inactive form[1].

  • Expression

Tissue_specificity:It is expressed (protein level) in neurons in the central region of the brain. It is expressed in the brain.

  • Isoforms & Post-Translational Modification

    Q03405 has 3 isomers: Q03405-1: 36978 Da (predicted); Q03405-2: 31263 Da (predicted); Q03405-3: 32016 Da (predicted).

  • Subunit

Monomer (Probable). Interacts with MRC2. Interacts (via the UPAR/Ly6 domains) with SRPX2. Interacts with FAP (seprase); the interaction occurs at the cell surface of invadopodia membrane. Interacts with SORL1 (via N-terminal ectodomain); this interaction decreases PLAUR internalization (PubMed:14764453, PubMed:23486467). The ternary complex composed of PLAUR-PLAU-SERPINE1 also interacts with SORL1 (PubMed:15053742). Interacts with CD82; this interaction prevents PLAUR from binding to its high affinity ligand PLAU (PubMed:15677461)
