TP-808 is a key enone intermediate for the discovery and manufacture of Tetracycline (HY-A0107) analogs and synthetic tetracyclines including eravacycline^[1].

In Vitro

TP-808 features highly modified stereocenters and functional groups (such as dimethylamino, TBS-protected phenoxy, benzyloxy, and isoxazole rings). It can rapidly construct the tetracyclic ABCD core skeleton of tetracyclines via Michael-Dieckmann condensation with a D-ring precursor, and serves as an essential building block for achieving "convergent total synthesis of tetracyclines and breaking through the bottleneck of semi-synthetic modification"^[1].

Protocol 1

Add each solvent one by one: 10% DMSO 90% Corn Oil

Solubility: ≥ 2.5 mg/mL (5.18 mM); Clear solution

This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown). If the continuous dosing period exceeds half a month, please choose this protocol carefully.

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL Corn oil, and mix evenly.
