Camylofin dihydrochloride
Camylofine dihydrochloride is an antispasmodic agent with myogenic and neurogenic effects. Camylofine dihydrochloride exhibits mild anticholinergic activity and blocks the M3 receptors on colonic smooth muscle. Camylofine dihydrochloride inhibits phosphodiesterase, elevates intracellular cAMP levels and reduces intracellular Ca2+ levels, thereby promoting smooth muscle relaxation. Camylofine dihydrochloride can be used in studies related to antispasmodia, gastrointestinal smooth muscle and ulcerative colitis.

In Vitro
The cumulative release rate of Camylofine dihydrochloride from Camylofine dihydrochloride-loaded Eudragit® S-100-coated chitosan microspheres reaches 72.5% in the presence of 3% w/v rat caecal content within 24 h, while it is 59.2% in the absence of rat caecal content[1].
After incubation for 24 h in a release system containing 3% w/v cecal contents from enzyme-induced rats treated for 5 days, the cumulative release rate of Camylofine dihydrochloride from Camylofine dihydrochloride-loaded Eudragit® S-100-coated chitosan microspheres reaches 95.5%[1].

In Vivo
Aqueous solution of Camylofine dihydrochloride (0.71 mg/kg; oral administration) reduces the Ca2+ concentration in colonic smooth muscle tissue to 1.989 mg/g colon in TNBS-induced colitis rats, whereas the concentration is 3.807 mg/g colon in the colitis control group[1].
