Background
14-3-3 zeta is an adapter protein involved in regulating a broad spectrum of general and specialized signaling pathways. It binds to numerous partner proteins, typically by recognizing phosphoserine or phosphothreonine motifs, often resulting in modulation of the partner's activity. This protein promotes cytoplasmic retention and inactivation of the TFEB transcription factor by binding to phosphorylated TFEB. Additionally, it enhances ARHGEF7 activity on RAC1 and facilitates lamellipodia and membrane ruffle formation. In neurons, 14-3-3 zeta regulates spine maturation through modulation of ARHGEF7 activity (By similar).
