BLNK, a central linker protein, operates downstream of the B-cell receptor (BCR), serving as a crucial bridge that connects the SYK kinase to various signaling pathways and regulates diverse biological outcomes in B-cell function and development. Its functional spectrum includes playing a role in the activation of ERK/EPHB2, MAP kinase p38, and JNK, as well as modulating AP1 activation. BLNK is pivotal for the activation of NF-kappa-B and NFAT, contributing significantly to BCR-mediated PLCG1 and PLCG2 activation, Ca(2+) mobilization, and the trafficking of the BCR to late endosomes. Notably, BLNK is dispensable for pre-BCR-mediated activation of MAP kinase and phosphatidyl-inositol 3 (PI3) kinase signaling. Additionally, it may be crucial for the RAC1-JNK pathway and plays a critical role in orchestrating the pro-B cell to pre-B cell transition.
