Background
DCP2 is a decapping metalloenzyme that catalyzes the cleavage of the cap structure on mRNAs. It removes the 7-methyl guanine cap structure from mRNA molecules, yielding a 5'-phosphorylated mRNA fragment and 7m-GDP. DCP2 is essential for mRNA degradation in both normal mRNA turnover and nonsense-mediated mRNA decay, and plays a role in replication-dependent histone mRNA degradation. It exhibits higher activity towards mRNAs lacking a poly(A) tail but shows no activity towards a cap structure without an RNA moiety. The presence of an N(6),2'-O-dimethyladenosine cap (m6A(m)) at the second transcribed position of mRNAs confers resistance to DCP2-mediated decapping. Under nutrient-rich conditions, DCP2 blocks autophagy by repressing the expression of ATG-related genes through degradation of their transcripts.
