Background

Cyclin D3 is a regulatory component of the cyclin D3-CDK4 (DC) complex that phosphorylates and inhibits retinoblastoma (RB) protein family members, including RB1, thereby regulating the cell cycle during G(1)/S transition. Phosphorylation of RB1 triggers the dissociation of the transcription factor E2F from the RB/E2F complex, leading to the transcription of E2F target genes essential for G(1) phase progression. Cyclin D3-CDK4 complexes serve as key integrators of mitogenic and antimitogenic signals. Additionally, Cyclin D3 forms part of the ternary complex cyclin D3/CDK4/CDKN1B, which is required for the nuclear translocation and activity of the cyclin D-CDK4 complex. Cyclin D3 also exhibits CDK4-independent transcriptional coactivator activity in collaboration with ATF5.
