Cyclin T1 is a regulatory subunit of the cyclin-dependent kinase pair (CDK9/cyclin-T1) complex, also known as positive transcription elongation factor B (P-TEFb). This complex facilitates the transition from abortive to productive elongation by phosphorylating the C-terminal domain (CTD) of the large subunit of RNA polymerase II (RNA Pol II). Cyclin T1 activates the protein kinase activity of CDK9 by mediating the formation of liquid-liquid phase separation (LLPS), enhancing the binding of P-TEFb to the CTD of RNA Pol II. During HIV or SIV infections, Cyclin T1 binds to the transactivation domain of the viral transcriptional activator Tat, increasing Tat's affinity for the transactivating response RNA element (TAR RNA). It serves as an essential cofactor for Tat, promoting RNA Pol II activation and enabling the transcription of viral genes.
