Description

CSK is a non-receptor tyrosine protein kinase that critically regulates cell growth, differentiation, migration and immune responses by phosphorylating tyrosine residues in the C-terminal tail of Src family kinases (SFK). This induces an inactive conformation through intramolecular interactions with the SH2 domain of SFK. CSK Protein, Human (Active, sf9, GST) is the recombinant human-derived CSK protein, expressed by Sf9 insect cells , with N-GST labeled tag.

Background

CSK, a non-receptor tyrosine-protein kinase, plays a crucial role in regulating cell growth, differentiation, migration, and immune response. It phosphorylates tyrosine residues within the C-terminal tails of Src-family kinases (SFKs), including LCK, SRC, HCK, FYN, LYN, CSK, or YES1. This phosphorylation triggers intramolecular interactions between the phosphotyrosine tail and the SH2 domain of SFKs, inducing an inactive conformation. To exert its inhibitory function on SFKs, CSK is recruited to the plasma membrane through interactions with transmembrane or adapter proteins in close proximity to the membrane. CSK effectively suppresses signaling from various surface receptors, such as the T-cell receptor (TCR) and B-cell receptor (BCR), by phosphorylating and maintaining several positive effectors, including FYN or LCK, in an inactive state.

The specific activity was determined to be >50 nmol/min/mg using Poly(Glu,Tyr) 4:1 as substrate.
