Background
RF1 is a component of the eRF1-eRF3-GTP ternary complex, which mediates translation termination in response to stop codons. The complex binds to a stop codon in the ribosomal A-site, with ETF1/ERF1 responsible for stop codon recognition and peptidyl-tRNA hydrolysis. Following GTP hydrolysis, eRF3 dissociates, allowing ETF1/eRF1 to fully occupy the A-site and facilitate peptidyl-tRNA hydrolysis. RF1 is also part of the transient SURF complex, recruiting UPF1 to stalled ribosomes during nonsense-mediated decay (NMD) of mRNAs containing premature stop codons. Additionally, RF1 is required for SHFL-mediated translation termination, which inhibits programmed ribosomal frameshifting (-1PRF) in viral and cellular mRNAs. by similar: Functional mechanisms of the eRF1-eRF3-GTP complex and its role in translation termination.
