Background

Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) exhibits dual functionality, serving as both a key enzyme in glycolysis, catalyzing the conversion of D-glyceraldehyde 3-phosphate (G3P) into 3-phospho-D-glyceroyl phosphate, and as a nitrosylase with roles in nuclear functions. In glycolysis, GAPDH plays a pivotal role in the initial step of the pathway. Beyond its metabolic functions, GAPDH modulates the organization and assembly of the cytoskeleton, facilitating microtubule and membrane associations through interaction with CHP1. Additionally, it is a component of the GAIT complex, participating in interferon-gamma-induced transcript-selective translation inhibition during inflammatory processes. In the realm of innate immunity, GAPDH contributes to TNF-induced NF-kappa-B activation and type I interferon production through interactions with TRAF2 and TRAF3, respectively. Furthermore, GAPDH is involved in various nuclear events, including transcription, RNA transport, DNA replication, and apoptosis, with its nitrosylase activity mediating cysteine S-nitrosylation of nuclear target proteins such as SIRT1, HDAC2, and PRKDC.
