Background
Glypican-2 (GPC2) Protein is a cell surface proteoglycan characterized by the presence of heparan sulfate. It may play a role in the motile behaviors of developing neurons. GPC2 interacts with Pleiotrophin (PTN) through its heparan sulfate side chains, promoting neurite outgrowth. This interaction involves the binding of PTN with chondroitin sulfate of proteoglycans, leading to the release of protein tyrosine phosphatase receptor sigma (PTPRS) from chondroitin sulfate proteoglycans (CSPGs) and subsequent binding with heparan sulfate of GPC2. Furthermore, GPC2 interacts with Midkine (MDK) through its heparan sulfate chain, inducing GPC2 clustering. This interaction is inhibited by heparin followed by chondroitin sulfate E and induces neuronal cell adhesion and neurite outgrowth.
