Background

gp41 antigen (gp41 antigen 2) is a transmembrane protein within the HIV envelope glycoprotein complex. By associating non-covalently with gp120 to form viral surface spikes, gp41 plays a critical role in mediating viral entry into host cells. The gp41 antigen exhibits high specificity; for instance, synthetic peptide antigens corresponding to specific regions of the gp41 glycoprotein encoded by the HIV-2 env gene have been developed. These specific antigens can elicit an immune response with HIV-2-specific antibodies and are widely used in diagnostic assays for HIV-2 infection as well as in vaccine research aimed at inducing antibody production in animals and humans. Furthermore, gp41 serves as a key target in HIV-1 immunology and vaccine design research. Its membrane-proximal external region (MPER) contains epitopes recognized by various broadly neutralizing antibodies (such as 2F5 and 4E10). Epitope scaffold technology can be employed to induce antibodies targeting specific gp41 conformations, thereby facilitating the analysis of viral virulence factors or the development of novel AIDS vaccines. In clinical antiviral therapy, gp41 is also the target of fusion inhibitors like enfuvirtide (T20), with mutations in its HR1 region serving as a critical determinant of drug sensitivity.
