Description

Background

IFI35 Protein acts as a signaling pathway regulator in the innate immune system response. In response to interferon IFN-alpha, it forms a complex with the transcriptional regulator NMI, preventing proteasome-mediated degradation of IFI35 and leading to IFI35 dephosphorylation. This complex also inhibits virus-triggered type I interferon/IFN-beta production and negatively regulates nuclear factor NF-kappa-B signaling. This inhibition results in the prevention of endothelial cell proliferation, migration, and re-endothelialization of injured arteries. Beyond its intracellular role, IFI35 functions extracellularly as damage-associated molecular patterns (DAMPs), promoting inflammation when released by macrophages during cell injury and pathogen invasion. Macrophage-secreted IFI35 activates NF-kappa-B signaling in adjacent macrophages through Toll-like receptor 4/TLR4 activation, inducing NF-kappa-B translocation and the release of pro-inflammatory cytokines. IFI35 homodimerizes and interacts with B-ATF, TRIM21, and directly with NMI in a complex facilitated by TRIM21.
