Background

IL-12 beta Protein, as known as IL12 p40 subunit or IL-12B, heterodimerizes with the IL-12 p35 subunit (IL-12A) to form IL-12 and with the IL23 p19 subunit to form IL-23, exerting different regulating functions[1].
IL-12 and IL23 belong to IL-12 family, are involved in proinflammatory responses and expressed by activated macrophages that serve as an essential inducer of Th1 cells development[2].
IL-12 signals via IL-12Rβ1 and IL-12Rβ2 mediated by p-STAT4, whereas IL-23 signals via IL-12Rβ1 and IL-23R mediated by p-STAT1 and p-STAT3[3].
The sequence of amino acids in IL-12 beta proteins of mouse is very different from human (69.04%), but shows high similarity with rat (92.24%).
Interleukin 12 (IL-12) family have been known to be inflammatory factors, induce autoimmune inflammation and thus may be responsible for autoimmune inflammatory diseases and may be important for tumorigenesis[4].

In Vitro

Mouse IL-12 (mIL-12) has a significant stimulating effect on CTLL-2 cells and results 65-7% of CTLL2 cell population having mIL12 receptors[5].
Mouse IL-12 (mIL-12) (.19-5 U/mL; 24-72 h) inhibits CTLL-2 cells proliferation without cell-lethal effect at 2 U/mL for 72 h[5].

In Vivo

IL12b-/- (p4-deficient) mice exhibits decreasing blood neutrophil numbers and an absence of serum levels of IL-17A[6].
  IL12b-/-/Itgb2-/- (lacking adhesion molecules like beta2-integrins) mice has a reducing tissue mRNA expression of IL-17A compared with Itgb2-/- controls and reduces total number of CD3+ IL-17A-producing Tn cells in the spleen and lamina propria[6].
IL12b contributes to Aβ42-induced neuronal damage and cognitive decline in APPPS1 Alzheimer's disease mouse model[7].
