It plays a role upstream of or within the negative regulation of interleukin-6 production. The protein is predicted to be located in both the cytoplasm and extracellular region, suggesting its potential involvement in diverse cellular processes. Furthermore, IL-36RN is anticipated to be active in the extracellular space. In humans, this gene is associated with pustular psoriasis 14, indicating its relevance in skin pathology. Notably, IL-36RN shares orthology with the human interleukin 36 receptor antagonist (IL36RN). Measured by its ability to inhibit IL-36 beta induced IL-6 secretion by NIH-3T3 mouse embryonic fibroblast cells. The ED50 for this effect is 0.7930-0.8070 μg/mL, corresponding to a specific activity is 1239.1574-1261.034 U/mg, in the presence of 15 ng/mL of Recombinant Mouse IL-36 beta/IL-1F8.
