Background
AURB-INCENP is a component of the chromosomal passenger complex (CPC), which acts as a key regulator of mitosis. The CPC complex plays essential roles at the centromere to ensure proper chromosome alignment and segregation and is required for chromatin-induced microtubule stabilization and spindle assembly. AURB-INCENP serves as a scaffold regulating CPC localization and activity: its C-terminus associates with AURKB or AURKC, the N-terminus binds BIRC5/survivin and CDCA8/borealin to tether the CPC to the inner centromere, while the microtubule-binding activity within the central SAH domain directs AURKB/C toward substrates near microtubules. The flexibility of the SAH domain is proposed to enable AURKB/C to track substrates on dynamic microtubules while maintaining CPC docking to static chromatin (by similarity). Additionally, AURB-INCENP activates AURKB and AURKC, regulates the localization of CBX5 to mitotic centromeres, and controls the kinetochore localization of BUB1.
