Background
The NA/Neuraminidase protein plays a crucial role in viral and cellular glycoconjugates by catalyzing the removal of terminal sialic acid residues. During virus budding, it facilitates the release of progeny virus by cleaving off terminal sialic acids on the glycosylated HA, preventing self-aggregation and ensuring efficient spread from cell to cell. Acting as a receptor-destroying enzyme, NA removes terminal sialic acid from cellular receptors, allowing the virus to spread through the circulation by further removing sialic acids from the cell surface. This activity is vital for avoiding limitations to one round of replication. Moreover, NA may enhance viral invasion of the upper airways by cleaving sialic acid moieties on airway epithelial cell mucin. Its association with lipid rafts during intracellular transport suggests a role in the budding process, potentially influencing host range restriction on replication and virulence. Notably, the sialidase activity within late endosome/lysosome traffic appears to enhance virus replication, highlighting the multifaceted functions of NA in the viral life cycle.
