Background

IRS-1 is a signaling adapter protein that mediates signal transduction from the insulin receptor (INSR) and insulin-like growth factor I receptor (IGF1R). It plays a critical role in development, growth, glucose homeostasis, and lipid metabolism. Upon phosphorylation by the insulin receptor, IRS-1 acts as a signaling scaffold, propagating insulin action by binding to SH2 domain-containing proteins such as the p85 regulatory subunit of PI3K, NCK1, NCK2, GRB2, or SHP2. Recruitment of GRB2 activates the guanine nucleotide exchange factor SOS1, triggering the Ras/Raf/MEK/MAPK signaling cascade (by similar). The PI3K/AKT pathway, activated by IRS-1, mediates most of insulin's metabolic effects, while the Ras/Raf/MEK/MAPK pathway regulates gene expression and, in coordination with PI3K, controls cell growth and differentiation. Additionally, IRS-1 positively regulates the Wnt/β-catenin signaling pathway by suppressing autophagy-mediated degradation of DVL2, thereby promoting cell proliferation.
