Background
MLK3 is a kinase that activates the JUN N-terminal pathway. It plays a crucial role in serum-stimulated cell proliferation and mediates the activation of MAPK14 (p38), MAPK3 (ERK), and MAPK8 (JNK1) by mitogens and cytokines through phosphorylation and activation of MAP2K4/MKK4 and MAP2K7/MKK7. Additionally, MLK3 is involved in mitogen-stimulated phosphorylation and activation of BRAF, though it does not directly phosphorylate BRAF. It also influences microtubule organization during the cell cycle.
