Description

NAMPT protein catalyzes the synthesis of nicotinamide mononucleotide, a key intermediate in NAD biosynthesis. NAMPT Protein, Human (sf9, His, GST) is the recombinant human-derived NAMPT protein, expressed by Sf9 insect cells, with N-His and N-GST labeled tag.

Background

NAMPT serves as the catalyst for the condensation reaction between nicotinamide and 5-phosphoribosyl-1-pyrophosphate, giving rise to nicotinamide mononucleotide—a pivotal intermediate in the biosynthesis of NAD. As the rate-limiting factor in the mammalian NAD biosynthetic pathway, NAMPT holds a central role in cellular processes. Beyond its enzymatic function, the secreted form of NAMPT exhibits dual properties, acting as both a cytokine with immunomodulating effects and an adipokine with anti-diabetic attributes. Notably, its extracellular function lacks enzymatic activity, partially due to the absence of activation by ATP, which is present at low levels in the extracellular space and plasma. Moreover, NAMPT plays a crucial role in the modulation of circadian clock function, orchestrating the oscillatory production of NAD to regulate clock target gene expression by facilitating the release of the CLOCK-BMAL1 heterodimer from NAD-dependent SIRT1-mediated suppression.
