TFAM Protein, Mouse (His, Myc)

Cat. No.: HY-P701316
COA Handling Instructions

TFAM proteins are critical in the regulation of mitochondrial transcription, bind to light chain promoters and are essential for basal mitochondrial DNA transcription. In the initiation complex of TFB2M and POLRMT, TFAM recruits POLRMT, and TFB2M induces structural changes in POLRMT, promoting promoter opening. TFAM Protein, Mouse (His, Myc) is the recombinant mouse-derived TFAM protein, expressed by E. coli , with N-10*His, C-Myc labeled tag. The total length of TFAM Protein, Mouse (His, Myc) is 201 a.a., with molecular weight of 30.9 kDa.

Background

The TFAM protein plays a pivotal role in mitochondrial transcription regulation by binding to the mitochondrial light strand promoter. As a crucial component of the mitochondrial transcription initiation complex, which includes TFB2M and POLRMT, TFAM is essential for the basal transcription of mitochondrial DNA. Within this complex, TFAM recruits POLRMT to a specific promoter, while TFB2M induces structural changes in POLRMT, facilitating promoter opening and trapping of the DNA non-template strand. TFAM is also indispensable for accurate and efficient promoter recognition by the mitochondrial RNA polymerase. It further promotes transcription initiation from the HSP1 and the light strand promoter by binding immediately upstream of transcriptional start sites. With the ability to unwind DNA and bend the mitochondrial light strand promoter DNA into a U-turn shape via its HMG boxes, TFAM plays a critical role in maintaining normal levels of mitochondrial DNA. Moreover, it may contribute to organizing and compacting mitochondrial DNA and potentially function in transcriptional activation or have a structural role in the compaction of nuclear DNA during spermatogenesis.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

  • Dilution Calculator

  • Specific Activity Calculator

The reconstitution calculator equation

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration
= ÷

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×
C1   V1   C2   V2

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
Unit/mg = 106 ÷ ng/mL
