Neutrophil cytosol factor 1 (NCF1) is an essential component, along with NCF2 and a membrane-bound cytochrome b558, for the activation of the latent NADPH oxidase, a process crucial for superoxide production. It forms an integral part of the NADPH oxidase complex, which includes the membrane proteins CYBA and CYBB, along with the cytosolic subunits NCF1, NCF2, and NCF4. NCF1 interacts with NCF2 through its C-terminus, forming a key association within the complex. Additionally, it engages with NCF4, CYBB, and CYBA, demonstrating its central role in coordinating the assembly of this complex. Interactions with NOXA1, ADAM15, TRAF4, and FASLG further emphasize the regulatory network of NCF1.
