Background
Nicastrin protein is an essential subunit of the gamma-secretase complex, an endoprotease assembly pivotal for catalyzing the intramembrane cleavage of integral membrane proteins, including Notch receptors and amyloid-beta precursor protein (APP). Operating in Notch and Wnt signaling cascades, the gamma-secretase complex regulates downstream processes by processing key regulatory proteins and modulating cytosolic CTNNB1 levels. As a fundamental component of the gamma-secretase complex, Nicastrin collaborates with a presenilin homodimer (PSEN1 or PSEN2), APH1 (APH1A or APH1B), and PSENEN/PEN2 to form the functional complex. This intricate molecular machinery binds to proteolytically processed C-terminal fragments C83 and C99 of APP and engages in interactions with PSEN1 and PSEN2, underscoring its significant role in molecular pathways and cellular processes.
