Background
PKN1 is a PKC-related serine/threonine-protein kinase involved in diverse biological processes, including regulation of actin cytoskeletal intermediate filaments, cell migration, tumor cell invasion, and transcriptional regulation. It functions within a signaling cascade initiated by adrenergic receptor ADRA1B activation, leading to MAPK14 activation. PKN1 modulates the cytoskeletal network by phosphorylating proteins such as VIM and neurofilament proteins NEFH, NEFL, and NEFM, inhibiting their polymerization. Additionally, it phosphorylates MAPT/Tau at Ser-575, Ser-637, and Ser-669, reducing its microtubule-binding affinity and disrupting tubulin assembly. PKN1 acts as a key coactivator of androgen receptor (AR)-dependent transcription by targeting AR-regulated genes and mediating phosphorylation of histone H3 at Thr-11 (H3T11ph), an epigenetic mark promoting KDM4C/JMJD2C-dependent demethylation of H3K9me. It also phosphorylates HDAC5, HDAC7, and HDAC9, impairing their nuclear import. Other substrates include PPP1R14A (Thr-38), MARCKS (Ser-159, Ser-163, Ser-170), GFAP, and in vitro, RPS6.
