RAB29 is a small GTPase that serves as a key regulator in vesicle trafficking. It is essential for maintaining the structural integrity of the endosome-trans-Golgi network (by similarity). In collaboration with LRRK2, RAB29 participates in the retromer-dependent retrograde trafficking pathway, recycling proteins such as the mannose 6 phosphate receptor (M6PR) between lysosomes and the Golgi apparatus. Additionally, RAB29 recruits LRRK2 to the Golgi complex and enhances its kinase activity, promoting LRRK2-mediated phosphorylation of RAB10 at Thr-73. Within the intact central nervous system (CNS), RAB29 regulates neuronal process morphology (by similarity). Studies also suggest a potential role for RAB29 in the formation of typhoid toxin transport intermediates during Salmonella enterica serovar Typhi (S. typhi) infection of epithelial cells.
