Background
SLC17A5 is a multifunctional anion transporter that operates via two distinct mechanisms: proton-coupled anion cotransport and membrane potential-dependent anion transport. It functions as an electroneutral proton-coupled acidic monosaccharide symporter with a 1:1 sugar-to-proton stoichiometry, exporting glucuronic acid and free sialic acid derived from sialoglycoconjugate degradation out of lysosomes, driven by the outwardly directed lysosomal pH gradient. This protein may regulate lysosome function and the metabolism of sialylated conjugates, impacting oligodendrocyte lineage differentiation and myelinogenesis in the central nervous system (by similar). Additionally, it acts as an electrogenic proton-coupled nitrate symporter, transporting nitrate ions across the basolateral membrane of salivary gland acinar cells with a 2:1 nitrate-to-proton stoichiometry, potentially contributing to nitrate clearance from serum by salivary glands. SLC17A5 also utilizes membrane potential to drive the uptake of acidic amino acids and peptides into synaptic vesicles, storing L-aspartate and L-glutamate in pinealocytes and facilitating vesicular uptake of the N-acetyl-L-aspartyl-L-glutamate neuropeptide, which is relevant to aspartatergic-associated glutamatergic neurotransmission and the activation of metabotropic receptors that inhibit subsequent transmitter release (by similar).
