Description

The T-PA protein encoded by this gene is a secreted serine protease that converts plasminogen into plasmin, a key enzyme involved in tissue remodeling and cell migration. The proteolytic process produces heavy and light chains, forming heterodimeric enzymes. T-PA Protein, Human (HEK293) is the recombinant human-derived T-PA protein, expressed by HEK293 , with tag free.

Background

T-PA Protein, encoded by this gene, serves as a secreted serine protease crucial for the conversion of the proenzyme plasminogen to plasmin, a key fibrinolytic enzyme involved in tissue remodeling and cell migration. The preproprotein undergoes proteolytic processing by plasmin or trypsin, generating heavy and light chains that associate via disulfide linkages to form the heterodimeric enzyme. T-PA's enzymatic activity plays a pivotal role in maintaining the delicate balance between fibrinolysis and coagulation. Increased activity leads to hyperfibrinolysis, resulting in excessive bleeding, while decreased activity can lead to hypofibrinolysis, potentially causing thrombosis or embolism. Alternative splicing produces multiple transcript variants, including at least one isoform that undergoes proteolytic processing. The gene exhibits broad expression, with notable levels observed in the urinary bladder (RPKM 61.1), placenta (RPKM 41.3), and 18 other tissues, reflecting its diverse roles in various physiological contexts.

Activité biologique

Measured by its ability to cleave a peptide substrate, Z-Gly-Gly-Arg-AMC. Read at excitation and emission wavelengths of 380 nm and 460 nm (top read). The specific activity is 3403.37 pmol/min/µg, as measured under the described conditions.

Formulation

Lyophilized from a 0.22 μm filtered solution of 50 mM Tris, 10 mM NaCl, pH 7.5.
Note: For SPR assay, please replace the buffer. Primary amine components (e.g., Tris, imidazole) can affect protein-coupled chips.

Endotoxin Level

<1 EU/μg, determined by LAL method.

Reconstitution

It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH2O. For long term storage it is recommended to add a carrier protein (0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose).

Storage & Stability

Stored at -20°C for 2 years from date of receipt. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.

Livraison

Room temperature in continental US; may vary elsewhere.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×
