Background

TAOK1 is a serine/threonine-protein kinase involved in multiple biological processes, including the p38/MAPK14 stress-activated MAPK cascade, DNA damage response, and cytoskeleton stability regulation. It activates the p38/MAPK14 cascade by phosphorylating MAP2K3 and MAP2K6, thereby participating in the G2/M checkpoint response to DNA damage. Additionally, TAOK1 phosphorylates Thr-208 of MARK2 to activate its kinase activity, leading to the detachment of MAPT/TAU from microtubules and maintaining cytoskeletal stability. This kinase also regulates apoptotic morphological changes (e.g., cell contraction, membrane blebbing) via the MAPK8/JNK cascade. TAOK1 plays an essential role in neuronal development within the central nervous system (PubMed:33565190) and is implicated in neuronal migration to the cortical plate (by similarity).
