Background

The TDGF1 Protein, a GPI-anchored cell membrane protein, emerges as a key participant in Nodal signaling. Functioning as a Nodal coreceptor in cis, cell-associated CRIPTO, when shed by TMEM8A, dynamically modulates Nodal signaling by enabling soluble CRIPTO to serve as a Nodal coreceptor on neighboring cells. This shedding mechanism contributes to the intricate regulation of Nodal signaling pathways. Moreover, TDGF1 is implicated in the determination of epiblastic cells, which subsequently give rise to the mesoderm, underlining its significance in early developmental processes.
