Background

TRPM4 is a calcium-activated selective cation channel that mediates membrane depolarization. While activated by increased intracellular Ca2+, it remains impermeable to Ca2+ itself. The channel transports monovalent cations (Na(+) > K(+) > Cs(+) > Li(+)), causing membrane depolarization and playing a central role in cardiomyocytes, entorhinal cortex neurons, dorsal root neurons, vomeronasal neurons, endocrine pancreas cells, kidney epithelial cells, cochlear hair cells, and other cell types. It participates in T-cell activation by modulating Ca2+ oscillations post T lymphocyte activation, which is required for NFAT-dependent IL2 production. TRPM4 is involved in myogenic constriction of cerebral arteries, controls insulin secretion in pancreatic beta-cells, and may contribute to pacemaking or irregular electrical activity during Ca2+ overload. The channel differentially regulates calcium signaling and NFATC1 localization to influence Th1/Th2 cell motility and cytokine production, while also enhancing cell proliferation via upregulation of the β-catenin pathway. Additionally, it plays a role in keratinocyte differentiation.
