Background

USP5, a deubiquitinating enzyme, demonstrates a marked preference for cleaving both linear and branched multiubiquitin polymers, with a particular affinity for branched structures. It is actively involved in the disassembly of unanchored 'Lys-48'-linked polyubiquitin chains. While USP5 binds linear and 'Lys-63'-linked polyubiquitin with a comparatively lower affinity, its knockdown results in the accumulation of p53/TP53 and an elevation in p53/TP53 transcriptional activity. This effect is attributed to the ability of the unanchored polyubiquitin accumulating in the absence of USP5 to compete with ubiquitinated p53/TP53, but not with MDM2, for recognition by the proteasome.

Verified Bioactivity

The fundamental role of USP5 is specific removal of ubiquitin from substrates. USP5 catalyses the ubiquitin from the substrate Ub-Rho110 to release fluorophores. Rho110 will release 535nM emission light under the excitation condition of 485nM. The signal of which can be quickly and reliably captured using a microplate reader.
