Background
Ku70 is a single-stranded DNA-dependent ATP-dependent helicase that plays a key role in DNA non-homologous end joining (NHEJ) by recruiting DNA-PK to DNA, participating in double-strand break repair and V(D)J recombination. It is also involved in chromosome translocation, preferentially binding to fork-like ends of double-stranded DNA in a cell cycle-dependent manner and working in the 3'-5' direction. During NHEJ, the XRCC5-XRRC6 dimer recognizes and binds to broken DNA ends, protecting them from further resection, and regulates DNA-PK complex activity by increasing PRKDC's affinity to DNA 100-fold. Ku70 may also function as a 5'-deoxyribose-5-phosphate lyase (5'-dRP lyase), cleaning abasic site termini. It acts as a negative transcriptional regulator with APEX1 and activates osteocalcin expression in association with NAA15. Additionally, Ku70 modulates DNA virus-mediated innate immune response by forming the HDP-RNP complex and suppresses apoptosis by interacting with BAX.
