Function:N6-methyladenosine (m6A) refers to the methylation modification of the sixth N atom of adenylate (A) in the base composition of RNA. It is the most abundant internal RNA modification in eukaryotes and one of the most common post-transcriptional modifications on RNAs, ubiquitously present in the RNA of living organisms from Escherichia coli to humans. It is found in most RNAs including mRNA, tRNA, rRNA, microRNA, snoRNA, etc., and is particularly prevalent in eukaryotic messenger RNAs (mRNAs). It is dynamic and reversible, catalyzed by RNA methyltransferases such as METTL3, METTL14, and WTAP (called "writers"), removed by demethylases such as fat mass and obesity-associated protein (FTO) and ALKBH5 (ALKB homolog 5) (called "erasers"), and recognized by m6A-binding proteins such as YTHDF domain-containing proteins and IGF2BPs (called "readers"). It participates in the regulation of gene expression through various mechanisms, including RNA alternative splicing, translocation, stability, translation efficiency, nuclear export, localization, and structural transformation, and is widely involved in processes such as cell proliferation, differentiation, embryonic development, and tumorigenesis. Its dysregulation is associated with the pathogenesis of diseases such as cancer, cardiovascular disease, and liver diseases (e.g., liver cancer, viral hepatitis, non-alcoholic fatty liver disease, hepatic fibrosis, acute liver injury, etc.).
