Description

Cyprinol (5α-Cyprinol) is an orally active bile salt and xenochemical pheromone. Cyprinol impairs renal function and increases plasma ALP activity in rats, induces diel vertical migration in Daphnia spp., enhances the permeability of the rectal mucosa to water-soluble compounds, and aids lipid digestion in fish. Cyprinol can be used in studies of fish toxicity and acute renal failure[1][2][3].

In Vitro

Cyprinol (1-140 mM; 30 min) exhibits hemolytic activity against sheep red blood cells, with an ED50 of 4.3 mM[2].
Cyprinol (25 mM; 15, 30 min) increases protein release from everted rat rectal sacs in vitro[2].

In Vivo

Cyprinol (100 mg/0.6 mL saline; p.o.; single dose) significantly impairs renal function and increases plasma ALP activity in male Wistar rats, but does not alter plasma AST or ALT activity[1].
Cyprinol (6.25-50 mM; rectal administration; single dose) potently enhances the permeability of water-soluble compound Ampicillin sodium (HY-B0522A) across rat rectal mucosa, with activity detectable at a concentration of 6.25 mM. Calcium ions do not inhibit its permeability-enhancing effect[2].
Cyprinol (10-10700 pM; aqueous exposure; continuous; 3-4 days) induces diel vertical migration in Daphnia magna[3].

Animal Model: Wistar (male, 230-270 g)[1]
Dosage: 100 mg
Administration: p.o.; single dose
Result: Reduced RBC count to 796 ×104 cells/dl, hematocrit to 47.6%, and hemoglobin to 13.6 g/dl.
Left WBC count unchanged at 5300 cells/dl relative to control.
Elevated plasma BUN to 16.7 mg/dl, plasma creatinine to 0.62 mg/dl, urine BUN to 462 mg/dl, and urine creatinine to 17.5 mg/dl significantly relative to control.
Left plasma AST (139 U/l) and ALT (47 U/l) unchanged relative to control.
Increased plasma ALP to 332 U/l significantly relative to control.
Animal Model: Wistar rats (male, 220-300 g)[2]
Dosage: 6.25 mM, 12.5 mM, 25 mM, 37.5 mM, 50 mM (co-administered with sodium ampicillin at 15 mg/kg)
Administration: rectal; single administration
Result: Significantly enhanced rectal absorption of AMP Na at 6.25 mM (blood AMP Na concentrations were detectable, unlike when AMP Na was administered alone).
Maintained nearly constant area under the concentration-time curve (AUC) of AMP Na at concentrations ≥12.5 mM, with peak blood AMP Na concentrations decreasing slightly at higher concentrations.
Further increased AMP Na absorption when co-administered with 25 mM calcium ion at 25 mM.
Significantly suppressed the absorption-enhancing effect when co-administered with 12.5 mM phosphatidylcholine dipalmitoyl at 25 mM.
Animal Model: Clone B (5-day-old neonates from second, third, or fourth clutches)[3]
Dosage: 1 pM, 10 pM, 107 pM, 1070 pM, 10700 pM
Administration: aqueous exposure; continuous; 3-4 days
Result: Induced a significant shift in daytime residence depth at concentrations ≥100 pM.
Produced mean daytime residence depth comparable to that induced by extracted fish incubation water, with statistically significant differences from control groups.
