Endo-β-N-acetylglucosaminidase D  (Synonyms: Endo D)

Endo-β-N-acetylglucosaminidase D (Endo D), isolated from Streptococcus pneumoniae. Endo-β-N-acetylglucosaminidase D hydrolyzes Fc N-glycan of intact IgG antibodies after sequential removal of the sialic acid, galactose, and internal GlcNAc residues in the N-glycan. Endo-β-N-acetylglucosaminidase D possesses transglycosylation activity with sugar oxazoline as the donor substrate^[1].

Protocol
1) For 10 μg of Fetuin, add 1 μL of 500 mM sodium acetate (pH 7.5), and adjust the volume to 10 μL with deionized water;
2) Add 1-2 μL Endo D, gently pipet to mix;
3) Incubate at 37 °C for 1 hour;
4) Analyze by SDS-PAGE or mass spectrometry.
Note: Incubation time or enzyme amount needs to be optimized according to actual conditions.
