Lulizumab (Humanized Anti-CD28 Recombinant Antibody) is an anti-CD28 domain antibody antagonist. Lulizumab inhibits T-cell activation by selectively targeting the CD28 signal. In a sensitized non-human primate kidney transplantation model, when combined with Carfilzomib (HY-10455), Lulizumab can regulate immune cells and prolong the survival time of the graft.

Animal Model:Male Macaca mulatta were sensitized by performing swapping skin transplants of 3cm diameter patches of dorsal skin between individuals of each pair twice, 8 weeks apart. Then they underwent kidney transplantation[2].

Administration:Subcutaneous injection; once a week for 4 weeks

Result:Led to a significant reduction in donor-specific antibody levels.
Decreased the number of follicular helper T cells and proliferating B cells in the lymph nodes.
Increased the proportion of naïve CD4 T cells and naïve B cells in circulation, maintained the frequency of regulatory T cells.
Significantly prolonged the median survival time of the grafts.
Caused animals to ultimately experience antibody-mediated rejection and graft failure.

Flow cytometric analysis of 106 Jurkat cells with Lulizumab (HY-P99302, red). Cells were fixed with 4% paraformaldehyde. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Human IgG H&L (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Cells without incubation with primary antibody were used as the unlabeled control (black).
